First of all you must be credible for the job you're applying. You cant apply to a computer programming job without knowing C++. And after you apply, don't sit and wait for them to call you, call them! Annoy the living crap out of them by calling them every couple of days so that you can get an interview and check your status: this way they will see that you're serious about getting the job and not just kinda a "well I'll fill out an application." Trust me, it works with every job I've gotten.

Volunteer at the place you want to work.
I did that over the summer and they saw how I could be a great help to them so I was hired right away. That way you learn if you enjoy it and you show them what you can do, with no obligations on either party.

Beer! .. again
Well not quite beer, but friends and socialising opens up new avenues, and more so if you have friends who are self employed or work for small to medium sized businesses - even if its not your dream job, its cash until you get something better.
I have gained a few jobs via friends in the past, the key is just dont be afraid to ask.

Persistence is the key. Don't get put off by a few rejections. Just apply for literally every job you are qualified for. Even then, it'll probably take you a few attempts at interviews before you know what to say to actually get the job.
